ARP-4791 contains the 3' long terminal repeat (LTR) region from an HIV-1 subtype E patient isolate upstream of the luciferase gene. The LTR was shown to be functional using the subtype B HIV-1, LAI, Tat protein. The LTR region (nt -147 to +63 relative to the transcription start site) from a subtype E patient isolate was PCR-amplified and exchanged into pBlue3'LTR-luc-B (BEI Resources ARP-4788) in place of the HIV-1, LAI, LTR. It contains two NFκB enhancers, three Sp1 binding sites, the TATA box and the TAR TNA hairpin motif. The construct is 5720 base pairs and is also referred to as pBlue3'LTR-luc-E.
