ARP-462 is a culture of WE17/10 cells, a mature human T lymphocyte derived from a patient with a slow-growing T-cell lymphoma. ARP-462 is interleukin-2 (IL-2)-dependent and express surface CD2, TCR/CD3, CD4, CD25, CD29, CD45RO, CD71, Leu8, and HLA class I and class II antigens.
ARP-462 does not contain integrated proviral sequences from known human retroviruses. The karyotype is abnormal [47, XX, -3, +der(3), t(1;3)(q41;q29) +M] in 100% of the cells.
Growth Characteristics: ARP-462 grows in a single cell suspension at high densities and in clumps at low cell densities. It is recommended that the cells be used at passage numbers below 50 for initiating long term infection experiments, and below 120 for other studies. This prevents the cells from acquiring characteristics resulting from long term tissue culture.
Growth Medium: RPMI 1640 medium supplemented with 20% fetal bovine serum, 10 mM HEPES, 30 to 100 units per mL human IL-2, 1.25 mM L-glutamine, 0.55 mM L-arginine HCl and 0.24 mM L-asparagine
ARP-462 is negative for Mycoplasma, bacteria and fungi.
Each vial of ARP-462 contains approximately 7 × 106 cells per mL in growth medium containing 10% dimethyl sulfoxide (DMSO). Please refer to the appropriate data sheet for lot-specific information.
