ARP-175 is a culture of MOLT-4 Cells, Clone 8, a human T-cell line originally derived from the peripheral blood of a 19-year-old male with acute lymphoblastic leukemia in relapse. Clone 8 was obtained by subcloning in soft agarose, as described in Kikukawa, R., et al. “Differential Susceptibility to the Acquired Immunodeficiency Syndrome Retrovirus in Cloned Cells of Human Leukemic T-Cell Line Molt-4.”J. Virol. 57 (1986): 1159-1162. PubMed: 2419583.
ARP-175 is highly susceptible to human T-cell lymphotropic virus III (HTLV-III). Following infection, cytopathic effect is observed after one day, and giant cells after five days. ARP-175 is a suitable host to isolate and continuously produce Simian immunodeficiency virus-infected African green monkey (SIVagm; ARP-174).
Growth Characteristics: Cells grow as a suspension. Maintain cultures at 3 × 105 per mL. Split cultures twice a week at 1:4 to 1:5.
Growth Medium: RPMI 1640 supplemented with 10% fetal bovine serum, 2 mM L-glutamine, 100 U per mL penicillin and 100 µg per mL streptomycin
Each vial of ARP-175 contains approximately 4.4 × 106 cells in crypreservative. Please refer to the appropriate data sheet for lot-specific information.
